The Leinster scrum-half has picked up a thigh injury and will miss the game with the knock.
His provincial teammate Luke McGrath has been called into the squad to provide cover.
It means a totally new half-back partnership for Ireland this weekend, with Johnny Sexton also ruled out through injury.
Joey Carbery and Conor Murray could be in line for starting roles.
