Seven tries against a porous Connacht defence sent Munster to the top of PRO12 table on the final day of the regular season.
Rassie Erasmus' side defeated the reigning champions 50-14 to set up a semi-final against Ospreys in Limerick. Keith Earls, Jack O'Donoghue, Andrew Conway (x2), James Cronin, Francis Saili and Conor Oliver all crossed the whitewash in a one-sided match in Thomond Park.
Connacht's two tries came from Cian Kelleher and Naulia Dawai. Pat Lam's side will now meet Northampton Saints in Franklin's Gardens in the Champions Cup Play-Off semi-finals. The winners will play either Cardiff Blues or Top 14 opposition in a view to playing in next season's competition.
Munster started the day three points behind Leinster at the top of the PRO12 table, but finished in first place as Ulster defeated Leinster by four points in Belfast.
Two tries from Roger Wilson and Andrew Trimble helped Ulster to a 17-13 win. Leinster will now play Scarlets in their semi-final, with a place in the PRO12 final at the Aviva Stadium at stake.
Leinster's only try came courtesy of a penalty try, as Kingspan Stadium said goodbye to Ruan Pienaar and Wilson with a win, to finish in fifth place in the table.
Scarlets' 40-17 win at home to Ospreys was also enough to ensure they finished in third place in the table.
In Italy, Treviso defeated Zebre 19-3 to book the sole Italian spot in next season's Champions Cup. Both teams started the final round level on points.
